The Fugu library provided by the Resource Centre was constructed by Elgar and Nizetic (unpublished data). It has an average insert size of of 40 Kb and consists of approximately 76,000 clones. The vector used is Lawrist 4. More information about the library may be obtained from the Genome News, 20 (1995).

The library contains 52,992 clones arrayed in 138 (384-well) plates.
The characteristics of the Fugu genome, along with the large evolutionary distance between bony fish and mammals make Fugu a useful tool for studying gene evolution.

How to find Fugu Cosmid clones
Find the genomic sequence of your region of interest from Ensembl and use the BLAST search at the link below, selecting the Fugu Cosmid ends from the database dropdown on the tool.
The search result shows lines connecting the Cosmid ends spanning the area of interest, http://www.fugu-sg.org/blast/.
The Cosmid end sequences can be found by clicking on the links in the allignment results.To find them on our website you have to remove the x1 or y1 and the Fc:c, and remove any 0s from the plate location after the letter, e.g. Fc:c032K19y1 becomes RC32-K19, Fc:c010C22y1 becomes RC10-C22.

Protocols / Clone Handling
Clones are streaked onto LB agar containing kanamycin (25µg/ml).
To use: restreak the clone onto the same agar type, in such a way as to obtain single colonies. Incubate at 37°C overnight.
Single colonies are then ready for DNA isolation, if required, using the accompanying protocol.
Clones can be grown in LB broth + kanamycin, and glycerol added after growth to a concentration of 25% for long term storage at -70°C.
